中文摘要: 以葡萄糖和甘油作为共底物培养基,将利用葡萄糖产甘油的酵母Candida krusei和利用甘油产1,3-丙二醇的细菌Klebsiella pneumoniae进行混菌培养,考查了双菌混合培养转化葡萄糖生成1,3-丙二醇的能力。首先考察了不同初始甘油和葡萄糖浓度对发酵结果的影响,在葡萄糖浓度为100 g/L和甘油浓度为80 g/L时得到较好的结果。然后在混合培养基中,对Candida krusei和Klebsiella pneumoniae分别独立培养时的生长情况,与双菌混合培养时的菌体生长情况作了比较,并同时对底物的利用和产物的生成情况作了比较。结果表明,葡萄糖的加入会抑制Klebsiella pneumoniae的生长,但是随着Candida krusei对葡萄糖的不断利用,葡萄糖对Klebsiella pneumoniae的生长的抑制会减弱。双菌混合培养可以将部分葡萄糖转化为1,3-丙二醇,但是其转化效率不高。

Abstract:Candida krusei which consumed glucose to produce glycerol and Klebsiella pneumoniae which consumed glycerol to produce 1,3-propanediol were co cultured in a co substrate medium composed of glucose and glycerol to study their ability of convering glucose to 1,3-propanediol. The effect of initial glucose and glycerol concentration on products concentration was investigated firstly. The results showed that higher 1,3-PD concentration was obtained at the initial glucose concentration of 100 g/L and initial glycerol concentration of 80 g/L. The growth curves of individual Candida krusei, Klebsiella pneumoniae and co culture in co substrate medium were compared, and correspondingly the glucose, glycerol, 1,3-PD concentration curves were compared. The results showed that the present of glucose in medium would inhibit the growth of Klebsiella pneumoniae, especially in individual culture in co substrate medium, but with the consumption of glucose by Candida krusei the inhibition function was decreasing. Co culture of Candida krusei and Klebsiella pneumoniae converted partial glucose into 1,3-PD, but the yield was lower and the correlation of Candida krusei and Klebsiella pneumoniae deserved advance research.
